Learning his wife may have survived a deadly inter-planetary war, a devoted husband's fight to find her becomes inspiration for one last stand against human extinction.

Maybe I'm less insightful than the others, but this worked for me. I'm not looking for a synopsis, just something to tweak my interest and it did (helps that I'm SF oriented).
I can see the protaganist (devoted husband). I can see the inciting incident (learned ... survived). I have an idea of the milieu (interplanetary war). I am told where its going (fight to find her, inspiration to others).
I can cope with the open questions of last stands and human extinction. But if you can make that connection better, go for it.
Has it been done before? Quite possibly but if you have a fresh version, well done. I notice "Godzilla" is about to come again. Like is that the 50th remake?
Happy writing.
Ditto what was said above.
The "fight to find her" could be expanded into the actual 1st goal/plan of the protag. For instance, maybe he is an engineer and builds a ship that can go to the alien world or fight the alien ships. Or, like Elysium, he builds a suit that makes him a super soldier. I think this is critical, explaining what the guy is doing to reach his goal.
Here's a take on your premise as an example, though it's a bit wordy.
"A scientist discovers the weapon used to vaporize humans in an interplanetary war is actually a transporter and assembles a team of survivors to liberate the enslaved humans, his wife among them, on the alien home world."
I had a couple of questions, It would be great to have a little more info about the couple, who are they, what do they do, are they soldiers? civilians? How recent was the war? What is the main obstacle he faces in finding her? Does his action inspire other people as your logline suggests? In which case who?
Where does this take place? On earth? In space? I think it might help if we had a tiny bit more specifics.
